學術(shù)與應用研究:

從事天然產(chǎn)物心血管藥理學活性與機制研究。重點方向是具有心血管藥理活性并具有重復結(jié)構(gòu)單元的天然產(chǎn)物多糖的化學結(jié)構(gòu)、結(jié)構(gòu)修飾、藥效活性、藥理機制及構(gòu)效關系研究。

巖藻糖化糖胺聚糖(fucosylated glycosaminoglycan, FG)結(jié)構(gòu)研究方面,基于bottom up策略,通過建立糖苷鍵選擇性解聚技術(shù)及寡糖純化與波譜學結(jié)構(gòu)解析技術(shù),闡明了一系列不同物種來源的天然FG的確切結(jié)構(gòu);獲得一系列用于藥理學活性與機制研究的、具有特殊端基結(jié)構(gòu)的純品FG寡糖及其結(jié)構(gòu)修飾產(chǎn)物。

系統(tǒng)研究了FG及其解聚產(chǎn)物、純品寡糖的抗凝抗血栓活性及其藥理學機制特點,重點研究了FG寡糖選擇性抑制內(nèi)源性因子X(Tenase, Xase)構(gòu)效關系、必須結(jié)構(gòu)、酶學動力學特點與機制、靶蛋白相互作用的動力學過程與機制。

基于天然FG結(jié)構(gòu)修飾,研制選擇性抑制內(nèi)源性Xase的新靶點抗凝抗血栓新藥LFG-53(First In Class,獲FDANMPA臨床許可),系列專利獲得國內(nèi)、美、日等國授權(quán),已實現(xiàn)知識產(chǎn)權(quán)及關鍵技術(shù)的企業(yè)技術(shù)轉(zhuǎn)移。

海王集團國家級企業(yè)技術(shù)中心工作期間,組建藥物研發(fā)實驗研究平臺,主管創(chuàng)新藥物研發(fā)。其代表性工作包括:立項并研發(fā)中藥天然藥物1類新藥虎杖苷注射液(臨床批件(現(xiàn)已取得FDA臨床許可))5類中藥新藥艾心酮片(臨床批件)、中藥大品種改進藥物鼻淵軟膠囊(上市許可)等。
